Testing and Verification for Combination Products
Upon completion of design output generation and formal design reviews, high-integrity testing and verification serve as the definitive technical proof of combination product device performance. Operating in alignment with FDA 21 CFR 820.30 and ISO 13485 standards, rigorous evidence is required to confirm that physical design outputs meet all technical design inputs. This phase follows a thorough framework that can be summarized in five stages.

Strategic Test Planning & Protocol Development
- Verification Master Plan (VMP) Authoring: Defining the overarching "rules of the game" for how every technical requirement will be challenged.
- Technical Test Protocol Writing: Developing step-by-step instructions with clear, procedural steps to challenge device's attribute functionality, and measurable Acceptance Criteria for each design input.
- Statistical Sample Size Justification: Applying rigorous statistical methods to ensure test results provide the high confidence levels required by ISO 16142 for medical safety.
Comprehensive Bench & Performance Testing
- Mechanical Characterisation: Executing stress, strain, fatigue, and high-cycle durability testing to identify physical failure points.
- Electrical Safety & EMC Testing: Coordinating leakage current, insulation, and electromagnetic compatibility studies per IEC 60601-1 standards.
- Software Unit & Integration Verification: Running formal code execution tests to ensure software outputs match design specifications without bugs.
- Performance Testing: The testing is performed against a matrix of testing criteria1 (cap removal force, activation force, injection time, dose accuracy, extended needle length, etc.) with validated methods, using statistically justified sample sizes, and with set acceptance criteria.
Biocompatibility & Environmental Validation
- ISO 10993 Compliance Testing: Managing material assessments for toxicity, irritation, and sensitisation to ensure safe human contact.
- Sterilisation Validation: Proving the efficacy of EtO, Autoclave, or Gamma methods in eliminating pathogens without compromising device integrity.
- Accelerated Shelf-Life & Stability Studies: Simulating years of storage in environmental chambers to verify device performance at the end of its labelled life.
Distribution & Transit Simulation
- Vibration & Drop Testing: Simulating the rigours of the global supply chain, including truck vibration and "worst-case" loading dock falls.
- Climatic Stress Testing: Exposing devices and packaging to extreme temperature and humidity fluctuations to ensure sterile barrier integrity.
Traceability, Reporting & DHF Integration
- Requirement Traceability Matrix (RTM) Management: Maintaining the master link between every Design Input, its specific Test Protocol, and the final Test Report.
- Deviation & Non-Conformance Management: Conducting root cause investigations for failed tests and managing the re-verification process after design iterations.
- Final Verification Summary Reporting: Compiling the definitive technical evidence required for the Design History File (DHF) and regulatory submissions.
